幾種疾病外周血單核細胞對

OK-432增殖反應(yīng)的比較

[摘要]　目的　研究掌跖膿皰病(pustulosispalmarisetplantaris,PPP)、尋常型銀屑病(psoriasisvulgaris,PV)、泛發(fā)性膿皰型銀屑病(generalizedpustularpsoriasis,GPP)、連續(xù)性肢端皮炎(acrodermatitiscontinua,AC)患者的外周血單個核細胞(peripheralbloodmononuclearcells,PBMC)對白介素-2(interleukin-2,IL-2)或鏈球菌粉干制劑OK-432的增殖反應(yīng),探討幾種疾病的關(guān)系。方法　分離提取PPP、GPP、PV、AC及健康患者PBMC,分別加入IL-2或OK-432,第6天MTT法檢測細胞增殖活力并測定DNA含量。結(jié)果　PPP、GPP、PV、AC患者與健康對照組的PBMC對IL-2的增殖反應(yīng)相比較無統(tǒng)計學(xué)差異(P>0.05)。而GPP、PV、AC患者的PBMC對OK-432呈低增殖反應(yīng),與健康對照組或PPP組有統(tǒng)計學(xué)差異(P<0.01)。PPP患者的PBMC對OK-432呈高增殖反應(yīng),與健康對照組無統(tǒng)計學(xué)差異(P>0.05)。結(jié)論　PPP和健康對照組對OK-432的反應(yīng)相似,而與其它幾種疾病反應(yīng)不同,提示PPP與其它幾種疾病可能存在不同的發(fā)病背景。

Abstract:Objective　Tofindthedifferentresponsesofperipheralbloodmononuclearcells(PBMC)tointerleukin-2(IL-2)orOK-432amongpustulosispalmarisetplantaris(PPP),psoriasisvulgaris(PV),generalizedpustularpsoriasis(GPP)andacrodermatitiscontinua(AC)invitroandtoexploretherela-tionshipsamongthediseasesabove.Methods　PBMCwereobtainedfromthesepatientsandhealthycontrols,thenIL-2orOK-432wasaddedinthemandtheproliferationactivityandDNAcontentwereas-sayedatthesixthday.Results　WhenthePBMCwerestimulatedwithIL-2,nosignificantdifferencewasfoundinproliferationresponsesbetweenpatientswithPPP,PV,GPP,ACandhealthycontrols.However,PBMCfromPV,GPP,andACpatientsshowedsignificantsmallerresponsestoOK-432thanthosefromthehealthycontrolsandPPP.Conclusion　ThedifferentresponsestoOK-432betweenPPPandPV,GPP,ACsuggestthatPPPhasadifferentpathogeneticbackgroundfromthatunderlyingPV,GPP,andAC.
